It is not necessary to obtain cut DNA samples to satisfactorily demonstrate the process. When using cut samples there is A requirement for A chemical, ethidium bromide, to highlight the DNA. This is A banned substance for use in schools - (Refer to the Ministry of Education manual Safety and Science revised edition page 57.) DNA Electrophoresis is routinely used as A means of separating fragments of DNA that differ in size. Samples are placed into wells at one end of an agarose gel, this is covered with A conducting buffer and an electrical current is passed across it. Because of its associated phosphate groups DNA has A net negative charge and will therefore naturally begin to migrate towards the anode. The agarose gel with its dense structure acts like A sieve, small fragments move easily through it but larger fragments, hindered by the gel, make slower progress. It is this differential speed of DNA pieces that cause them to separate out from each other. The dyes in the kit also migrate at variable speeds and can therefore be used to mimic the behaviour of different sized DNA fragments. The dyes that travel the greatest distance represent small pieces of DNA. The sample that make little progress are representative of larger pieces.
